RESUMO

ObjectiveTo study the genetic diversity and genetic relationship of Pinellia ternata germplasm resources and provide the basis for germplasm identification, variety breeding, and resource conservation. MethodIn this study, 27 P. ternata were used as experimental materials to determine seven phenotypic characters, such as plant height, leaf length, and leaf width. Simple sequence repeats (SSR) primers were designed based on P. ternata transcriptome data, and polymerase chain reaction (PCR) amplification was performed on 27 P. ternata samples. The genetic diversity of P. ternata germplasm was analyzed by POPGENE32, PowerMarker V3.25, and NTSYS-PC 2.10e software. ResultA total of 10 pairs of highly polymorphic primers (PIC>0.5) and four pairs of moderately polymorphic primers (0.25<PIC<0.5) were selected. The average number of alleles detected was 3.928 6, and the average Nei's diversity index (H) and Shannon's index (I) were 0.557 8 and 1.002 9, respectively, indicating a high level of genetic diversity. Cluster analysis divided the Pinellia ternata into seven categories, and P. ternata in the same province were in the same categories. The SSR molecular ID cards of 27 P. ternata germplasm were constructed with 14 pairs of primers, and the rapid identification of P. ternata in each region was realized. ConclusionThe results of this study can lay a foundation for the genetic diversity and population structure of P. ternata and provide a scientific basis for the identification of P. ternata germplasm resources, map construction, and molecular-assisted breeding.

Objective@#To investigate the effect of allogeneic red blood cell transfusion on T lymphocyte subsets and natural killer (NK) cells in peripheral blood of patients with gastric cancer during perioperative period.@*Methods@#50 patients with gastric cancer in our hospital from January 2017 to December 2017 were randomly divided into control group (n=25) and observation group (n=25). Perioperative allogeneic erythrocyte transfusion was performed in the observation group, while no allogeneic erythrocyte transfusion was performed in the control group. Blood samples were taken from two groups before and after operation 3 d, and serum T lymphocyte subsets (CD3+, CD4+, CD8+, CD4+ /CD8+ ) and NK cell levels were measured. T cell subsets and NK cell levels of patients in observation group, control group and observation group at different clinical stages before and after operation were counted. According to blood transfusion volume, the patients in the observation group were divided into <4 U group and <4 U group. T cell subsets and NK cell levels before and after operation were compared between the two groups.@*Results@#There was no significant difference in CD3+, CD4+, CD8+, CD4+ /CD8+, NK cells between the two groups before operation (P>0.05). The levels of CD3+, CD4+, CD4+ /CD8+, NK cells in the two groups after operation were lower than those before operation and CD8+ levels were higher than those before operation. The levels of CD3+, CD4+, CD4+ /CD8+, NK cells in the observation group were lower than those in the control group, and the levels of CD8+ in the observation group were higher than those in the control group (P<0.05). The levels of CD3+, CD4+, CD8+, CD4+ /CD8+, NK cells in patients with stage Ⅰ-Ⅱ and stage Ⅲ-Ⅳ had no significant difference before operation (P>0.05). After operation, the levels of CD3+, CD4+, CD4+ /CD8+, NK cells in the two groups were lower and CD8+ levels were higher than those before operation. The levels of CD3+, CD4+, CD4+ /CD8+, NK cells in stage Ⅰ-Ⅱ patients were higher than those in stage Ⅲ-Ⅳ patients, and CD8+ levels were lower than those in stage Ⅲ-Ⅳ patients (P< 0.05); There was no significant difference in the levels of CD3+, CD4+, CD8+, CD4+ /CD8+, NK cells between the <4 U group and <4 U group before operation (P>0.05). After operation, the levels of CD8+ in the two groups were higher than those before operation, while the levels of CD3+, CD4+, CD4+ /CD8+, NK cells were lower than those before operation; The levels of CD3+, CD4+, CD4+ /CD8+, NK cells in the <4 U group were higher than those in >4 U group, and CD8+ levels were lower than those in >4 U group (P<0.05).@*Conclusions@#Perioperative allogeneic red blood cell transfusion can affect T cell subsets and NK cell content in peripheral blood of patients with gastric cancer, and there are some differences in the effects of different clinical stages of gastric cancer patients, which can cause the suppression of cellular immune function.

Objective:To optimize the extraction technology and evaluate antioxidant activity of total flavonoids from Hedyotis dif-fusa. Methods:The content of kaempferol was detected by an HPLC method,and spectrophotometry was used to detect the content of total flavonoids. Results: The optimum extraction conditions were investigated by orthogonal design with the extraction quantities of kaempferol and total flavonoids as the evaluation indices. The antioxidant activities of the extracts were detected based on the clearance of hydroxyl radical and DPPH·model. The optimum extraction conditions were as follows:10-fold amount of 80% methanol and with ultrasonic extraction for 15 min. The clearance rate of 1ml methanol extract for scavenging DPPH· and ·OH was 73.89% and 91.27%,respectively.Conclusion:The optimum technology is stable and feasible for the extraction of Hedyotis diffuse. The extract shows good in vitro antioxidant properties, which provides powerful reference for the future development of relevant antioxidant prod-ucts.

Objective:To learn the depression status of the hospitalized pregnant women in labor and its related influencing factors,and to provide the theoretical basis for exploring depression intervention of the pregnant women in labor.Methods:Self-Rating Depression Scale was used for investigating the depression status of 203 hospitalized pregnant women in labor who were in hospital from November 2014 to May 2015,and the impacts of demographic characteristics,maternal history and physical condition and health care on depression status of hospitalized pregnant women in labor before pregnancy and during pregnancy were analyzed using Logistic regression.Results:Fifty-three people with depressive tendencies were detected among pregnant women in labor,and the positive rate was 26.1%.The differences in positive rates of depression tendency of the pregnant women between less than 30 years,30-39 years and 40 years and above had no statistical significance (P>0.05).Univariate Logistic analysis showed that the differences of occupation,overweight or obesity,low-dose folic acid supplements before pregnancy were statistically significant (P<0.01).Multivariate Logistic regression analysis showed that risk factor for pregnant women was overweight or obesity pre-pregnancy,and the risk was 2.335 times to non-overweight or non-obesity pregnancy women (OR=2.335,95%CI:1.101-4.954,P<0.05).Conclusion:The pregnant women have the depression tendency,especially the pregnant women with high body mass index before pregnancy.The knowledge of psychological health education should be provided for them.

Objective To investigate the effects and probable mechanism ofShenfu injection on the oxidaitve damage of H2O2-induced PC12 cells.Methods PC12 cells were cultured and exposed to 100μmol/L H2O2 for 1 h to establish the oxidative damage model. The protective effect ofShenfu injection was observed by the cell survival rate measured by colorimetric MTT assay, and the leakage rate of lactic dehydrogense (LDH). Western blot methods were used to detect the expression of NF-κB signaling pathway.Results Compared with the model group,Shenfu injection at 5, 10, 20 ml/L could improve the PC12 cells survival rate (83.11% ± 2.59 %, 87.99% ± 0.59%, 85.26% ± 1.07%vs. 73.82% ± 1.82%;P<0.01 orP<0.05), decrease the LDH leakage rate (32.75% ± 4.10%, 28.52% ± 1.14%, 35.79% ± 1.62%vs. 64.34% ± 3.18%;P<0.01 or P<0.05). Western blot results showed thatShenfu injection could protect the PC12 cells from oxidaitve damage by suppressing the p-p65/p65 (1.30 ± 0.10, 1.17 ± 0.06, 1.37 ± 0.15 vs. 1.70 ± 0.10;P<0.01 orP<0.05), p-IκBα/IκBα (1.07 ± 0.12, 1.00 ± 0.10, 1.03 ± 0.06 vs. 1.17 ± 0.06; P<0.01 orP<0.05).ConclusionShenfu injection has a obvious antioxidant effect on PC12 cells in vitro.

Aim To investigate the effect of indirubin derivative PHⅡ-7 and TRAIL on proliferation in breast cancer cell MCF-7 and its MDR counterpart MCF-7/ADR and the mechanism.Methods Growth inhibition rate was examined respectively by MTT assay under treatment with TRAIL or PHⅡ-7 or in combination. Cell apoptosis and ROS production were examined by flow cytometry.The change of TRAIL receptors(DR4/DR5 )in mRNA was analysed by realtime PCR.Re-sults IC50 of PHⅡ-7 on MCF-7 and MCF-7/ADR was (4.49 ±1.55 ),(3.44 ±0.90 )μmol · L-1 respec-tively;MDA-MB-231 was TRAIL sensitive cell line, and apparently TRAIL induced apoptosis in MDA-MB-23 1 .Low concentration of PHⅡ-7 in combination with TRAIL could augment TRAIL-induced cytotoxic effect including apoptosis while TRAIL or PHⅡ-7 treatment alone had limited cytotoxity to those cells.Besides, PHⅡ-7 at this concentration had little toxicity to hu-man peripheral blood mononuclear cells even if in com-bination with TRAIL.PHⅡ-7 generated ROS produc-tion inside MCF-7 and MCF-7/ADR cells and up-regu-lated DR4/DR5 expression concentration dependently. Once upon ROS scavenger NAC involved,the effect of TRAIL receptors up-regualtion by expression was abro-gated.Conclusions PHⅡ-7 at low concentration could improve the sensitivities of breast cancer cell MCF-7 and MCF-7/ADR to TRAIL,the mechanism of which may be the ability of ROS production by PHⅡ-7 help up-regulated TRAIL receptor DR4,DR5 .Our re-search set a solid foundation for PHⅡ-7 in combination with TRAIL in future clinical application.

Objective To analyze the expressions of osteopontin (OPN)and chemokine (CXC-subfamily)receptor 4 (CXCR4)in different ovarian tissues,and to explore the role of OPN and CXCR4 in occurrence,development and metastasis of human ovarian cancer.Methods The expressions of OPN and CXCR4 in normal ovarian tissue (n=20),benign ovarian epithelial tumor (n=20)and epithelial ovarian cancers tissues (n=40)were detected by immunohistochemical SP method,and the expression rates of OPN and CXCR4 in epithelial ovarian cancer tissue with different clinical characteristics were analyzed.Results The positive expressions rate of OPN in epithelial ovarian cancer tissue was significantly higher than those in normal ovarian tissue and benign ovarian tumor tissue, and there were significant differences in the positive expression rates of OPN between different pathological stages (G1 ,G2 ,G3 )of epithelial ovarian cancer (P 0.05).There was no positive expression of CXCR4 protein in normal ovarian tissue and benign ovarian tumor tissues , but there was positive expression in epithelial ovarian cancer tissue.Besides,the expressions of CXCR4 protein were not significantly different among different pathological grades,different clinical stages and different histological types (P > 0.05).Conclusion The OPN and CXCR4 expression levels are correlated with the degrees of malignancy in epithelial ovarian tumor,therefore the CXCR4 and OPN expression pathways may be the new targets for ovarian cancer therapy.

ObjectiveTo explore the application of peripherally inserted central venous catheter (PICC)and central venous catheter (CVC) in the surgical parenteral nutrition.MethodThe operation time,success rate,indwelling duration,abnormal biopsy,and adverse reactions of PICC and CVC were retrospectively compared.ResultsCVC had significantly shorter operation time than PICC groups [ (28.67 ±5.13) min vs.(45.20 ±6.89)min,P =0.035 ).The mean indwelling duration was significantly longer in PICC group than in CVC group [ ( 114.85 ± 10.29) d vs.(24.78 ± 8.42) d,P =0.033 ].PICC group was superior to CVC group in terms of five complication items,while only phlebitis was more frequent/severe in CVC group.ConclusionsPICC may be more suitable for long-term ( >4 weeks) catheter-based nutrition.CVC has short operation time and fast flow rate,and therefore is most suitable for rescue treatment for critically ill patients.

The clinical ability of employees in laboratory medicine refers to their ability to give useful information and helps for clinical diagnosis and treatment.In laboratory medicine education,the cultivation of the undergraduates'clinical ability should be strengthened.Case teaching and clinical signs analysis is practical to the cultivation of clinical ability.In addition,we should carry out the reform of the teaching materials in order to adapt to the requirements of the clinical ability training. Improving the clinical ability should be the students' a lifelong career planning.

Objective To investigate the expression and clinical significance of platelet-derived growth factor A (PDGF-A) on placenta tissue from pre-eclampsia.Methods The expression of PDGF-A in the placenta of 38 pre-eclampsia patients and 22 normal pregnant women at third trimester was detected by immunohistochemistry method.Results (1)PDGF-A was mainly expressed in the cytomembrane and cytoplasm of cytotrophoblasts and the endothelial cell of capillary in placenta.(2) The rates of PDGF-A expression of cytotrophoblasts were 63% (24/38) in pre-eclampsia group and 32% (7/22) in normal pregnancy group,which exhibited significant difference (P <0.05).(3) The rates of PDGF-A expression of endothelial cell were 68% (26/38) in pre-eclampsia group and 27% (6/22) in normal pregnancy group,which also showed significant difference (P <0.01).(4)The rates of PDGF-A expression of cytotrophoblasts were 39% (7/18) in mild pre-eclampsia patients and 85% (17/20) in severe pre-eclampsia,which reached statistical difference (P < 0.01).Conclusion The increasing expression of PDGF-A in cytotrophoblast and endothelial cell in placenta might confer the occurrence and progression of preeclampsia.

Objective To determine the relationship between acceptance of disability and posttraumatic stress response in patients with brachial plexus injury. Methods Total 160 patients with brachial plexus injury were recruited and investigated with the Impact of Event Scale-Revised (IES-R) and the Acceptance of Disability Scale (ADS). Results The total score of ADS was (79.07±11.99) which showed medium level of acceptance of disability. The total score of IES-R was 4-66 (33.51±14.41), which showed that most of the patients suffered from posttranmatic stress response. Significantly negative correlation was found between acceptance of disability and posttraumatie stress response(r=-0.480, P<0.001). Conclusions Nurses should pay more attention to the acceptance of disability and posttranmatic stress response in patients with brachial plexus injury, and provide appropriate health education and effective psychological intervention to improve the patients' mental health and quality of llfe.

Spiramycin and midecamycin are 16-membered macrolide antibiotics with very similar chemical structures. Spiramycin has three components, namely spiramycin I, II and III. Spiramycin II and III are, respectively, the O-acetyl and propionyl derivatives at C3-hydroxyl group of spiramycin I. Midecamycin has four components, and the C3-hydroxyl group of midecamycin is all O-propionylated. The enzyme adding acyl group(s) at the C3-hydroxyl group during the biosynthesis of spiramycin and midecamycin is 3-O-acyltransferase. The 3-O-acyltransferases for spiramycin and midecamycin are also very similar, and presume to function when exchanged. To explore whether the 3-O-acyltransferase for midecamycin biosynthesis hold still the character of selective and efficient propionylation for spiramycin I at its C3-hydroxyl group, we inserted mdmB, the 3-O-acyltransferase gene from Streptomyces mycarofaciens ATCC 21454 for midecamycin biosynthesis, into a mutant strain of S. spiramyceticus F21, in which the 3-O-acyltransferase gene for spiramycin biosynthesis, sspA, was deleted; and the mdmB was integrated exactly into the chromosomal site where the sspA was deleted. We name this "hybrid" strain as SP-mdmB. HPLC analysis of the spiramycin produced by SP-mdmB showed that spiramycin I was still the major component, although the relative proportions of both spiramycin II and III increased significantly. We thus conclude that MdmB from Streptomyces mycarofaciens ATCC 21454 for midecamyicn biosynthesis do not hold the character of selective and efficient propionylation for spiramycin I within S. spiramyceticus F21, and this character is possibly limited in Streptomyces mycarofaciens ATCC 21454 for midecamycin biosynthesis.

Case teaching method was used to undergraduates’specialized courses of clinical laboratory medicine such as the clinical laboratory hemotology and basic clinical laboratory medicine. This teaching method achieved satisfactory effect in clinical ability culture and obtained the positive opinion of students.
